Abstract:

Rare blood disorders encompass a diverse group of hematological conditions characterized by distinct pathological features, clinical presentations, and diagnostic challenges. This review aims to provide a comprehensive overview of the hematopathology of rare blood disorders, highlighting the key diagnostic considerations and recent advancements in understanding these conditions.

The first section of the review focuses on rare inherited hematological disorders, such as Fanconi anemia, paroxysmal nocturnal hemoglobinuria (PNH), and hereditary spherocytosis. For each disorder, we discuss the underlying genetic mutations, pathophysiology, and characteristic hematopathological findings, including peripheral blood smear, bone marrow morphology, and flow cytometry analysis.

The second section delves into rare acquired hematological disorders, including various types of thrombotic microangiopathies (TMA), such as atypical hemolytic uremic syndrome (aHUS) and thrombotic thrombocytopenic purpura (TTP). We explore the underlying mechanisms, such as complement dysregulation and ADAMTS13 deficiency, and their impact on hematopathology.
